Get gelatin from a reputable vendor. Usually proteins are soluble in weakly acidic water (0.1 N HCl in autoclaved ddH2O), prepare solution fresh, cover plasticware with gelatin for several hours, remove solution with sterile pipet, let it dry and expose plasticware to UV light under sterile culture hood (laminal flow) for a few hours until dry. Pack in Sarah Wrap or plastic and store at 4 degree Celsiu until use.

This is the recipe we use for gelatin for HUVEC that has been used in our lab for the last 15 years at least. We autoclave it after its made.

Sterile EF water (Baxter) 750 ml

Gelatin granular 15.39g (Sigma cat# G9391)

FCS 32.6 ml

Sterile 7.5% Na Bicarb. 19.6 ml

Sterile 10x HBSS with glucose 65.2 ml
