I am currently working with recently purchased ME-1 cells. I was wondering if anyone that has experience transfecting this cell line, or even lines that are similar, would be willing to share which methods they employed.
ME-1 cells are human Acute Myeloid Leukemia cells isolated from peripheral blood (Article Establishment and characterization of a new human leukemia c...
), they are cultured suspended in RPMI + FBS 20%.Early transfection experiments showed little to no transfection with Lipofectamine 3000 (at least without removing FBS from cells).
Lonza Nucleofection with a reporter gene (GFP) also has disappointing efficiency.

Thank you for your replies. I was hoping to find an alternative to lentiviral transfection, since i want to insert mutated oncogenes so security procedures are more complex.
Unfortunately these cells do not like being cultured alone, so selection with puromycin (my current approach) is not too helpful if transfection does not reach a certain minimum efficiency level.
I will look into FACS selection for my upcoming experiment though. Also, I am still open to other suggestions!
