according to my opinion 4°C should be enough for short period of time, for longer storage I would recommend -18 or -24°C.

I have two more issues:

1. I wanted to store the DNA pellet. I thought about air-drying the pellet and then

storing it. Now I read about storing the DNA pellet in 70% or 75% ethanol. What is the advantage of storing the pellet in ethanol compared to the air-dried pellet?

2. Is there really no problem with storing genomic DNA pellets at -20°C? As for genomic DNA in solution, I do not want to store it at -20°C as it might fragment upon freezing and thawing.

is it necessary to storage the DNA as a pellet?

For my purposes I storage DNA in high concentration solution (in H20 or TE) in -24°C (depend on the sample the concentration range from 100

to 500 uM/ul for DNA extracted with the application of CTAB method). That DNA solutions are thewed when there is a need to make working solution of 20 uM/ul. The quality of storaged DNA solutions are checked by electrophoresis and we did not record traces of degradation. I hope that it will be helpful.

I want to store the DNA as a pellet because when I process big amounts of DNA, I always have a problem with the time. That's why I want to store DNA pellets before I can continue purifying my DNA the next day (or a few days later).

I remember adding ethanol to DNA pellets and storing it at -20 C; it worked perfectly (no fragmentation later or anything). Unfortunately, I don't know why it is better than just a pellet.

It depends on the storage lenght and the quality of the DNA. Pelleted DNA could be saved for years at room temperature. You can also adsorb your DNA on sterile filter paper and store properly for years at room temperature too.

DNA pellet can sustain without loosing its quality at minimal 4 oC temperature for longer storage.
