I've been discussing this with a few people, but I'm seeking more definitive arguments. If one is looking to find whether a difference exists in Cytokine levels between cases and controls and also wants to profile the response (i.e. not just interested in one or two cytokines) multiplex is a good tool, but there are issues like somewhat lower sensitivity.
Questions are: What is the best way to deal with out of range values (below threshold) as opinions vary on this one? Are there implications for picking up small changes from baseline in cytokines that are present in extremely low concentrations in normals? Are there any other issues that would suggest that the use of ELISA vs multiplex might be a better option. Any input is appreciated as current opinions are quite diverse.
