I am optimising the TransIT LT1 reagent for CHO-DP12 cells . I have only achieved an efficacy of 6%. Does anyone have experience with this cell line and reagent and what protocol and efficacy did ye achieve.
I've used and compared TransIT to other reagents for CHO transfection. TransIT never worked well, hardly worked at all.
Polyethyleneimine (PEI) is a cheap and good reagent for CHO transfection with a DNA plasmid. Transfection efficiency should range between 40-50%, and the internet is full of protocols, and transfection volumes are scalable. Incubation of DNA and PEI for complex formation is not necessary, look for protocols without the incubation step, and save yourself some time.
