Hi all, I want to use flow cytometry selectively isolation vascular endothelial cells from tissue in inflammation state. It should exclude blood cells (such as hematopoietic cell, circulating endothelial progenitor cell). I already have the cdh5-ert mice. So basing cdh5 marker, which markers should I combine to isolate the vascular endothelial cells? Some paper use CD45-, some use antibody complex (blood lineage).

Which tissue are you interested in? For lymph nodes and tertiary lymphoid structures, you can stain for CD45, CD31 and gp38 (also known as podoplanin). You should gate on CD45-, then CD31/gp38. Fibroblastic reticular cells (FRCs) (gp38+CD31-) are the major stromal subset in lymph nodes, then blood endothelial cells (BECs; CD31+ gp38-) and LECs (gp38+CD31+).
