Hello,
Excluding contamination and probe degradation, can anyone suggest a cause of late amplification in the NTC of TaqMan qPCR ? I considered the possibility of primer dimers, but since they are not specific to the probe, they should not give any signal. Are there other types of secondary structure (in either the primers or probe) that could be causing this?

Hi,
is your NTC showing a linear rise or a curve shape? Linear ris can also origin from probe hydrolysis, some probes and dyes tend to show this

- At what point is your NTC signal at Ct
- If coming up in late 30 cycles so no Ct calculated you have no need to worry
- Indeed if your NTC is > 35 cycles and your actual signal ct is
