I've recently upgraded from the Agilent Seahorse XF24 to the XFe24 and have started having issues with the OCR calibration. The rep suggested that storing the hydrated cartridge at 4C even within 72 hours is potentially damaging the probes and messing with the calibration, so to keep it in the 37C incubator even over 24 hours.
Has anyone had issues like this? I'm nervous about what's going to grow in the wells over the weekend (and of course I don't want the cartridge to dry out!).

The recommendation to store the hydrated cartridge at 37°C for 72 hours is to avoid damage to the probes or calibration difficulties. However, holding the cartridge at 37 degrees Celsius for a lengthy period of time might result in the growth of undesired bacteria in the wells.
It is critical to follow the manufacturer's storage and maintenance instructions. To reduce the danger of contamination, some feasible methods include doing the OCR calibration soon after hydration or using a sterile procedure while handling the cartridge. Additionally, you might want to think about utilizing a microplate reader or an other sort of cartridge to measure OCR.

You should be able to use the plate if hydrated a minimum of 4 hrs. We have stored Seahorse cartridges in calibrant solution for up to 96 hrs at 28C (not in sterile conditions) without a problem. I haven't tried this at 37C though. After adding calibrant solution and reassembling the plate, we put the cartridge back in its box and tape its foil lid back onto it. This protects it from light and helps prevent drying out over a longer period of time.
Melissa Chernick Thanks for your input! The 37C worked, but the RT storage (with your mods :) ) might help keep contamination at bay.
