Hi!

I am working with reverse genetics derived influenza A virus (PR8), but I am having this issue for a quite a long time now!

After producing the working virus in MDCK and titering by plaque assay I infect A549 cells with a MOI of 3 but I only get around 40% of infected cells, when I theoretically (and by others experience) expected an infectivity of around 90%.

Do you have any ideia why it is happening?

I already produced new viruses (more than once) using new solutions of every component (BSA, trypsin TPCK).

Also, my infection media for the experiments in A549 is SFM DMEM with pen/strep 1%.
