The best one I've used is pCpGl luciferase reporter assay. It uses a CpG-less plasmid, meaning anything you clone into it and methylate with SssI allows you to ONLY study your promoter of interest. It's also a pretty straightforward luciferase assay and is very robust. See attached paper!
