I'm trying to remove dead cells from human Mononuclear cell culture without either slow centrifugation or ficoll.
I would like to remove dead cells by pulvevrising them into debris (smaller than RBC) which can then be removed via filtration.
I'll test Dnase next week, but aside from that, are there any other enzymes or solutions that can digest dead cells to debris whilst keeping live cells intact?
I believe you can use a cell sorting system (or a similar cytometric system in which you can detect size/granulation) to separate living cells from the dead cells if you have access to such equipment.
You may also try to find a detergent which in certain doses do not harm your living cells. You may check this paper to get an insight: http://www.pnas.org/content/100/10/5795. If you can choose such a detergent and determine a harmless concentration, I recommend you to treat your cells and observe if it had any effect on the dead cells. Usually it is harder to damage living cells as they have mechanisms to defend their structural integrity. Therefore it is safe to assume that there will be a concentration in which you can get rid of the dead cells without harming the living ones.
