Hi, I am working on experiments for finding cancer cell death signaling pathway induced by chemical co-treatment.
I find that co-treatment generates enough cell death, measured by PI staining on FACS.
This cell death was reduced by Z-VAD, but in Western blotting, There were no changes in caspase-3 or -8, -9 and PARP
And Autophagy inhibitor, like 3-MA, enhaced this cell death. I think this means that autophagy contribute to cancer-survival pathway.
If anyone went through results like this, Please give me any opinion.

We already know that autophagy enhances survival and prevents aging to some extend, which is possibly the reason the autophagy inhibitors in your experiments are showing increased death rate. My assays with PC3 cells, had same results. Caspase inhibitors would logically promote apoptosis the reason of your results, I guess.

PS- More interesting would be to explore how the malignant cells evade apoptosis to promote survival.
Very interesting, but why? Why cancer cells are more sensitive to autophagy than normal ones? Possibly due to ROS damage which makes them more available for autophagy attack.

I have only found about autophagy or apoptosis, but couldn't think about evasion of apoptosis in cancer.

Yes, actually I thought that probably ROS induces autophagy, because the chemical I use makes ROS in early times.
so I did experiment using ROS inhibitor, but results were, only DPI reduce cell death but not much, and other ROS inhibitors, like NAC, didn't work or enhance cell death.
