We have the cryopreserved form of Plasmodium beghei ANKA stain in liquid nitrogen vapour in our lab. We are for the first time going to inoculate the mice with the stain to induce rodent malaria model. We have prepared the protocol but I have few queries regarding the same.

1) After thawing at 37oC, can we directly inoculate the strain in to the mice or first we need wash it with normal saline or phosphate buffered saline (pH 7.4) to remove the cryoprotectant? As in the case of P. falciparum we use to wash the strain after thawing to remove the cryoprotectant before proceed to in vitro culture.

2) After inoculation to the mice, shall we need to keep the animals in separate room to prevent the transmission by vector mosquito to other mice or rodents?
