I have not used a fluorescence reader, but regardless of that, I am pretty sure with 100rpm you cannot suspend cells in wells and that'll probably impact your reads. I think it takes a couple trial and error.

Hi Hamid Kian Gaikani Thank you for your response, however, it seemed 100rpm (constant shaking) worked for me, atleast for growth but the fluorescence readings were not good.

cells grow even without shaking at even at cold temperature. but higher rpm is needed to suspend them well and have consistent reads.

I agree with that! However, I could not verify that because the machine stopped working and I switched to another one with 180 rpm.
