I got a flask of Caco-2 to expand, and they were growing well using DMEM High with 20% FBS. I freezed them at ~1 mi. cells/mL of freezing solution (90% FBS, 10% DMSO), kept it at -80°C for a day and then, at -196⁰C.
Now, I'm having trouble thawing them. I tried in a T25, with and without centrifuging; in a 6 wells plate; changing the medium in the day after, and changing the medium only after 72 hrs, and I'm still not getting it.
The cells seem to grow, and not necessarily are dying, but just a very small part of them are adhered to the flask. Most of it is in the supernatant, but it seems that some cells are partially adhered, in a way that they are discarded when I change the medium but they are kinda adhered.
Anyways, I don't know if it was because the way I freezed them, or if there's other tips to making it work.
