When ever I mount the cover slips on the slides to perform the microscopy I find crystals on the other side of the cover-slip after drying. please suggest me the better recipe where i can use it for both cell culture and microscopy.
10X PBS/1L I make 1X from this.
1.44g Na2HPO4 (10mM)
80g NaCl (1.37M)
2g Kcl (27mM)
2g KH2PO4 (18mM)

Also. you can prepare a 10X solution autoclaved and just prepare 450 ml H2O autoclaved in bottles. At the time of use you can simply add 50ml of 10X PBS and use it at the time of requirement and store the 1X PBS at 4 deg C. This is how it is managed in my cell culture laboratory. But I have no idea about microscopy slide preparation.

I think it is normal that you find crystals after drying. PBS is composed of salt and when water evaporate they will precepitate

The only way to remedy this is to follow up your PBS washes with ddH2O washes. Any form of PBS will leave salt crystals behind.

The problem is not the recipe but rather the procedure. PBS evaporating off the cover slip will leave salt behind. The cover slips need to be rinsed with ultrapure water.
