Hello All!

I want to transfect PLB 985 cells with quite big plasmids (around 12 kbp) and I am wondering which method will be the best.

I already tried nucleofection with Lonza 4D system set for the HL-60 program (worked perfectly with RNPs, over 90% efficiency), but cells die. Probably due to the plasmid size (I managed to sort few single-cell clones plates but with no edits, efficiency was less than 1%).

That is why I am considering other methods like Lipofectamine LTX with Plus (for hard-to-transfect cells) or a lentiviral system.

Does anyone have some hints?

I assume HL-60 experience with big plasmid transfection will be valuable too :)
