Hi all,
I am about to start culturing THP-1 cells, and am looking for suggestion regarding the addition of antibiotics (pen/strepto) to the culture medium (which should be: RPMI 1640 + 2mM L-glutamine + 10mM HEPES + 0.05 2-ME + 10%FBS).
I have found different protocols, with or without antibiotics.
What is your experience with that?
Anything else regarding the culture medium and the propagation conditions will be very much appreciated as well!

Most in our lab routinely use a mixture of antibiotics and antimycotics. Although we do have one researcher that does not, for fear that low grade infection (masked and kept in check by the antibiotics) may alter readouts of important immune stimulation and signaling. You just have to be very careful and fastidious with your aseptic techniques. We also use heat inactivated serum due to the fear that complement factors within the serum may activate the cells. Hi Sudip, thank you very much for replying. I decided to put penicillin(100U/ml)-streptomycin(100ug/ml) to my medium. But I will avoid amphotericin since it looks a bit harsh to me.
