Can anyone share their transfection protocol for NIH 3T3 cells. I tried with lipo3000 protocol. Very few cells are transfected and their shapes change after transfection.

NIH 3T3 cells are quite a pain in the neck to transfect with lipofectamine.
This has been extensively discussed in the forum.
The highest you can achieve is 40 to 50 % with transfection reagent. The best available transfection reagent is the NIH3T3 Transfection Kit available from Altogen Biosystems
Or, the other option is to go for electroporation.
