I'm having an issue with newly acquired MC3T3-E1 Subclone 4 cells adhering to my cell culture flasks. I am using BD Falcon (#353136) T75 flasks, and I am following the ATCC recommendations for media and supplements (alpha-MEM + 10% FBS + 1X anti/anti (Pen/strep/amphotericinB)) and culture conditions.

Upon thawing the original vial received from ATCC, I seeded the cells in a T25 flask where they proliferated, reached confluence, and I then transferred them to a T75 flask. The cells reached confluence again in the T75 flask, yet after my last passage (1:6) they failed to adhere. I spun them down once again and combined them into a 1 T25 (different brand- Corning) hoping that increased cell density and different brand flask may help. Unfortunately, they have not adhered nor proliferated.

Any suggestions as to what might be the problem, or recommendations for brand/type of cell culture flasks for MC3T3-E1? I have contacted ATCC and am awaiting a reply.
