Which markers can be used to define Mouse Memory B cell, I have got panel of CD19, B220, CD38, CD80,CD273 anitbody based on references, but now I am confused how to define Memory B cells from this panel.
I believe it may be ideal to include IgD and another immunoglobulin such as IgM in your panel. Unless you are excluding other cell types that express B220 and CD80 such as DCs, you may have some false artifacts in your "memory B cell" population. Adding the IgD and IgM helps to isolate only B cells which are producing those surface bound antibodies. The memory B cells would then be those which are IgD negative and IgM positive. However, immature B cells also have the same IgD- and IgM+ phenotype so you may have to add something that excludes immature B cells.
Your panel should work - the literature shows very heterogeneous populations of 'memory' B cells depending on the surface phenotype and the experimental setting. I think you've chosen the right panel to give you an idea of where your memory cells sit relative to other B cells that shouldn't express some of those markers
It seems to define Memory B cells, you most of all have to rely on the B220. That is because other mentioned AB in the panel are more specialized, such as: the CD80 defines functionally distinct memory B cell subsets that are independent of antibody isotype; the CD38 protein is a marker of cell activation; the CD19 gene encodes a cell surface molecule that assembles with the antigen receptor of B lymphocytes in order to decrease the threshold for antigen receptor-dependent stimulation; and the CD273 functions to inhibit T-cell proliferation by blocking cell cycle progression and cytokine production, and is particularly important in the maintenance of self tolerance.
