Does anyone know what the contaminant in this nanodrop image could be? Its from a phenol/chloroform/isoamyl extraction of a single mosquito larva. I washed twice with an excess of chloroform. The ethanol precipitation was carried out using 100 ul of aqueous layer in 0.3 M sodium acetate and 1 ul polyacryl carrier in over 1 ml of 100% ethanol. The pellet was washed once in 70% ethanol and was a light grey color. I resuspended in 20 ul of water. Thanks.
It may be easier to troubleshoot if you share the entire extraction protocol. For example, what is the pH of your phenol/chlor/iso? Is there an initial proteinase K treatment? However, one easy step would be to do one or multiple chloroform washes and an ethanol precipitation to remove whatever contaminant is present. Specific protocols are online, but basically you wash your final product with chloroform and re-precipitate with 100% cold ethanol and NaOAc overnight. You'll lose some product, but it cleans away contaminants like phenol. You may have also not let the ethanol evaporate completely before you added the elution buffer.
