I'm trying to extract the White Blood cells from Blood for cell culture. 

I'm currently using LSM to get the mononuclear cells but I think it would be easier just to lyse the red cells directly with something like ACK lysing buffer.

I've used ACK on mouse RBC's but we only use those cells for flow. I'm not sure of how viable the white cells are after lysing.

What are the advantages and disadvantages of using ACK over LSM?

I haven't been able to find anyone who's made any direct comparisons and I'd be nice to see if anyone knows before I try it
