My opinion try to draw the primer to amplify a specific region of 1 kb. If you try to amplify this big product your efficiency decrease much.

The efficiency of the qPCR is connected at more factors.For example to the Tm of the primers, concentration of DNA, concentration of primers, number of cycles, inhibitors inside the DNA samples.
