We want to transfect pCDH plasmid or other plasmids to suspension ExpiCHO-S cells and express our target proteins, and we used the transfection reagents PEI. Unfortunately, there is no eGFP expression and low transfection efficiency. Is there some effective reagents or methods? thanks a lot.

Have you tried lipofectamine 2000 (I think it is applicable to cells in suspension) or nucleofection (e.g. lonza amaxa or equivalent) ?

Recommended using pcDNA3.4 TOPO TA vector to get the best expression effect.
ExpiCHO-S cells should be passaged at a density of 3–4 x 106 cells/mL the day before transfection in order to reach a density of approximately 7–10 x 106 cells/mL on the day of transfection.
Before transfection, dilute the cells with fresh medium to a final density of 6 x 106 cells/mL and gently shake to mix.
The remaining cells should be discarded; do not continue to inoculate and culture with high-density cells.
