I'm working on cloning and expression of growth hormone receptor in mammalian system and biological activity (Binding activity) of growth hormone with its respective extracellular growth hormone receptor. My concerned question is, either the half life of this complex (Growth hormone and extracellular growth hormone receptor) in the absence/presence of GH will increase.
In order to check the sustained half life, I just want to predict it with some software before starting this experimental procedure.
The query is interesting and need utmost attention before execution. I will try to put my idea in this regard, but am not sure if it can meet your expectation. If you have the experimental structural complex information, then just go for few nanosecond MD simulation. Now calculate the BE using MMPBSA/MMGBSA. You can simulate the complex in the absence/presence of GH as you have mentioned with careful confrontational alternation inspection. Regarding half-life test, am not sure if you will be able to trace ant disintegration during few nanosecond as per AA-MD is concern. Still you can look into the CG-MD with few microsecond analysis.
