I am looking for the way to keep cell not dried after taking out of the PBS.
I measured the sample without coverlip but also not dried before.
When we are using coverslip, we used manicure to seal the coverslip preventing from drying.
Does anyone know how to make the cell sample by sealing the cell without coverslip?

This is a method that has worked for me to prevent cell samples from drying. You can use a suitable mounting medium to preserve your cells. After placing your sample on a slide, add a drop or two of the mounting medium, making sure to cover the entire area. Then, place a coverslip on top.
I recommend conducting some tests to optimize this method for your specific needs.
Hello, I have used the method of Gelled glycerol, commonly known as glycergel, is used as a mounting medium in microscopy techniques and sample preparation for observation. Glycergel is a transparent and viscous medium that helps preserve samples and prevent deterioration during microscopic observation. Here is an overview of how to prepare and use a glycergel mounting medium:
Preparation of mounting medium with glycergel:
Dilute the glycergel in a suitable buffer (eg, PBS OR 0.9% saline sol) to the desired concentration. The typical concentration is 50% to 70% glycergel in buffer and 0.1 to 0.5 phenol crystals as preservative.
Heat the diluted glycergel solution to approximately 50-60 °C to facilitate its dissolution and homogenization.
Gently mix the glycergel solution to ensure it is well dissolved and free of bubbles.
Allow the glycergel solution to cool to room temperature before using it.
Mounting of samples:
Prepare your samples according to the corresponding fixation and staining protocol.
Place a small amount of the glycergel solution on a clean, dry slide.
Place the sample in the glycergel solution, making sure to cover it completely.
Place a coverslip over the sample carefully to avoid the formation of air bubbles.
Make sure that the specimen is completely submerged in the glycergel mounting medium and that no bubbles are trapped.
Sealing and storage:
Seal the edges of the coverslip with clear nail polish or mounting sealer to prevent evaporation of the mounting medium and dehydration of the specimen (This is not necessary if the specimen will be used before 3-6 months of age, which is how long it takes in dehydrate).
Store mounted slides in a cool, dark place to protect samples from photodegradation (depends on your stain, if you used antigen and/or antibody retrieval).
The glycergel mounting medium provides a suitable environment for microscopic observation, maintaining the integrity of the samples and minimizing the fading of fluorescent signals.
