I have made up 5nm,10nm and 20nm paclitaxel in culture medium and stored at 4 degrees. I have also made up 0.25, 0.5 and 0.75 micro molar camptothecin in normal culture medium, stored at 4 degrees. I am concerned about the stability and efficacy of the drugs in culture medium over time.
We store paclitaxel and topotecan in DMSO frozen at -20 degrees celsius and thaw for use with no problems. For your reference.
In my case, I usually store the stock solutions of above compounds (10 mM) at -20 degrees celsius as Lynn Huang mentioned. The compounds are diluted in culture medium just before treatment.
Yong is correct. its always best to store a "super stock" which is 100x more concentrated than the concentration you want in cell cultures. DMSO is fine for camptothecin, I do not remember if we used Cremophor to dissolve the stock paclitaxel, but we definitely used Cremophor to dilute the super-stock to the final working solution of taxol--and it worked v. well. So, pl. check on Solubility of paclitaxel in cremophor versus DMSO and other organic solvents used for cell culture.
