CCL20 is a chemokine that works in tandem with MMP12. These two molecules are secreted simultaneously: the first ensures the attraction and the second allows migration.
Monocytes can produce large quantities of both molecules simultaneously. This requires that these monocytes are activated: a double activation is required: first the cells are cultured five days in GM + IL4, then these MODC are activated in the presence of IL10 + TNFa. Secretion of CCL20 is then between 1,000 and 10,000 times higher than that of PBL.
Most tumor cell lines can produce CCL20, but at a basal level.
If we compare the human and murine sequences of CCL20 are very close, however, the active region is rather variant. I'm not sure that human CCL20 may be perfectly functional on murine HEV, for example.
