We are currently working on a cytosolic protein; however, we think that this protein also has a secreted form and can be detected in the bloodstream. As there is no ELISA kit available for this protein, I was wondering what method could be used to detect its presence in mouse serum. Western blot would be one choice. Is there any better method to detect this protein in serum sample?
P.S. We do have antibody for this protein.
A Western blot would be the simplest approach. You could improve the sensitivity by using an albumin-removal resin to reduce the amount of albumin protein in the serum sample. You could also combine immunoprecipitation with Western blotting to further improve the sensitivity.
Another method is surface plasmon resonance, which requires specialized equipment. The antibody is immobilized on the sensor surface, and a signal is generated when the antigen binds to it.
