I'm trying to use new organic compounds as a remedy for sperm membrane injury, I want to use lysosomes as delivering vehicles? can any one give me an easy method for liposome preparation?
Various methods can be used according to what you need. There is a good review. Pharmaceutica Acta Helvetiae 70, no. 2 (1995): 95-111. You can see that, and some citing papers.
Hello Mr. Palani. Liposomes are very easy to prepare using the "Bangham method" or "hydration of thin lipid film". It depends on your lipids, molecule to be encapsulated, and other factors.
Bangham method:
-Dissolve lipids and hydrohobic molecules in organic solvent
-Evaporate organic solvent (rotatory evaporation) to form a thin lipid film inside a glass flask
-Hydrate the thin lipid film with an aqueous solution (may include hydrophilic compounds) at temperatures above the Tc of the main lipid of the thin lipid film
-Sonicate, extrude or employ other techniques to reduce size
-Separate liposomes with encapsulated drug from free drug (centrifugation for example)
-Store liposomes at 4°C
