Is there a way/protocol for changing the medium for mammospheres in culture?

Hi, I am currently using the breast cancer cell line JIMT-1 to generate mammospheres in culture, and have managed to see mammospheres in both primary and secondary generation. However I have noticed that the mammospheres in secondary generation appear alot smaller in size, and so I would like to continue growing these in culture.

My question is whether there is a way/protocol for changing the medium for mammospheres in culture? Currently I have just been adding 3mL of fresh MammoCult Medium every 3-5 days, however I'm using a 6-well low-attachment plate and so the maximum amount of medium I could put in there is 10mL.
