I've been isolating marine Streptomyces for about 15 yrs, but just switched to soil Strep. I typically use oatmeal agar to get these microbes to sporulate. I make spore stocks then for storage. Does anyone know if adding MgCl2 or CaCl2 to the media will help with the extra hard ones? I've tried ISP2, ISP4, oatmeal, R2YE and sporulation agar, but in some cases the microbes won't sporulate on any of those. I've read somewhere in the past that you can use either of them to revive a culture from a slant. If you have any media ideas I could try that would be greatly appreciated as well.
