Anyone knows of a HeLa cell line that expresses GFP under HIV-1 activated promoter? I'm trying to find a flow-based infectivity assay that would be comparable to HeLa TZM blue assay, with Gal or Luc as a readout. I see a Ghost cell line with GFP that's induced by HIV, but not sure how the result compares to TZMbl cells, due to a possible difference in surface expression of CD4/CCR5/CXCR4. Thoughts?

Yes, the HOS-derived GHOST cell lines are an excellent set of reporter lines. We have used these routinely. Produced by Dan Littman's group, there are a range of GHOST cells, transduced with the various coreceptors and CD4 (see https://www.aidsreagent.org/ and search for "GHOST"). Be aware that with increasing passage the proportion of cells with inducible GFP may decline. Make a large stock of these cells for liquid nitrogen storage when you first get them. Don't carry them too long in culture. The Littman lab have also provided extensive information on the culture and use of these cells.
