I'm measuring p-NDRG1 and NDRG1 in mouse tissues. While I have good confidence in my p-NDRG1 antibody (Cell Signalling #5482), I have little confidence in my total NDRG1 antibody (Cell Signalling #9408).
The variability I see in NDRG1 seems unlikely to represent 'real' biology (a beta actin loading control shows very little variability), and I question whether the band/s I'm quantifying are even really NDRG1.
There are several other commercial NDRG1 antibodies available (http://www.cellsignal.com/browse/?Ntk=Products&N=4294956287&Ntt=NDRG1), but one of my colleagues say they're all just as bad as #9408. I'd prefer to test that myself, but don't want to waste $1K and a weekend finding out he's right.
Does anyone have a good, working antibody? If so, can you share the product number and your recommended protocol (how much protein to load, how to block--Milk or BSA, dilution to use, etc.)?
