We are working with a recombinant peptide of 3 kDa, but we do not have tricine in our lab, only glycine. Is that possible to detect this peptide without using tricine in the SDS-PAGE? High concentrations of bis-acrylamide are enough to detect? Thanks!
I don't think you will be able to accomplish it with a standard tris-glycine, even with a very high percentage gel, such as 20%. There are some publications on special methods for peptide separation by gel electrophoresis, such as these:

If you have an HPLC or UPLC in your lab, you can do the analysis by reverse-phase chromatography instead.
Well, that depends on your sample. Do you expect to have other proteins with molecular weights below 10 kDa? If yes, you need the tricine system to resolve the low molecular weight proteins.
