In our lab, we use 35 mm plastic dishes with a 10 mm hole in the bottom, where we attach 22x22 mm coverslips using silica glue. We use this to grow cells and use it as the imaging dish for live cells. For immunofluorescence staining, we perform it on the same dish and use 1X PBS as an imaging medium.
In routine coverslip mounting protocol, removing the coverslip and placing it cell-side down onto a drop of mounting medium on a glass slide is mentioned. But the explants I culture would dislodge from the coverslip if I do so.
Has anyone ever performed mounting a sample, where the medium is directly put on top of the dish and sealed using another coverslip from the top?
