I understand that most RNA isolation kits, like QIAamp minikit cannot be used for isolating RNA from frozen samples. Are there any kits which may be used? what about manual methos with trizol
A collaborator was attempting to do this but was getting poor results. We helped them modify their protocol to include a step of RBC lysis and then isolation of PBMCs before freezing, and her RNA yields have increased significantly. Is there any DMSO in the blood prep to help prevent the cells from rupturing during the freeze/thaw? Here is another forum where they describe the best approaches for RNA isolation but many start with fresh blood samples. Thank you Shana, thats what we are doing now, isolating WBC before freezing but i am trying to look for protocol by which i can salvage some of the blood we froze earlier.
