Currently i am expressing a particular MMP in cell line (vero). signelling sequence of mmp was removed. The vector was having CMV promoter and transfected using PEI which was showing more than 50% of transfection with GFP alone. when i was trying to analyse the expressed protein by westernblot, i coudn't get band and moreover, i am using Santa cruze primary and secondary antibody. The primary antibody was rised against human mmp but we are using Dog mmp for our work. where is the problem in the technique and wat may be the reason for not getting band.
