I spreaded E.coli(TG1) that had infected by phage on agar plate and it was incubated at 30°C.
After about 15h, I didn't found colonies!
Afer 20h, I found some colonies(it seems like that it much grow) which were at the edge of plate. But overall, still rare colony....
I wonder if it is possible that E. coli can grow very slowly.. above 24h on agar plate

Hello Yeo-jin. May I know why you choose to incubate at 30'C? Have you tried incubation at the standard 37'C?
It is quiet possible that a particular type of E.coli grows slowly. I have used E.coli T7 Shuffle cell for protein expression which I used to incubate at 30°C. It took almost 24 hours to see some visible colonies. Incubation up to 30-36 hrs gives characteristics colonies of E.coli. You have mentioned that bacterium was infected by phages. Please let me know do you have any control plate which was not infected by phage. Depending on the comparison you can explain what is actually happening.
Are your phages lytic phages?
If not, TG1 cells grow very quickly and should grow at both 30 and 37 degrees (faster at 37, of course). Anyway, why do you consider that your colonies (obtained after 20h) are rare? Abnormal morphology?
