To determine whether changes in cell cycle are related to DNA damaged, we investigated the effect of our drug on cell cycle distribution by flow cytometric analysis. also the tested drug, triggers apoptosis in our cells. should we have expect to see sub-G1and G2 arrest at the same time?

DNA damage causes activation of p53 protein. The expression of p53 is necessary in cell cycle arrest, repair, and apoptosis. There are two checkpoints in the cell cycle which involves p53. The G1/S checkpoint is entirely p53-dependent, and the G2/M checkpoint which is p53-independent and can be accomplished because of multiple pathways.
When p53 gene is mutated G1/S checkpoint is lost and the cells no longer arrest at G1/S, but the arrest at G2/M is maintained because p53 only contributes to the maintenance of G2/M checkpoint as this checkpoint is p53-independent.
So, it is possible to see sub-G1and G2 arrest at the same time provided p53 protein is not mutated.
