so, basically i had to do an anti-fungal assay using a plant extract which was extracted using methanol. then i dissolved the extract in methanol to bring 200mg/ml working concentration. I made this working concentration by dissolving 200mg of the crude with 1ml methanol. Is it ok to use methanol or is it necessary to change the solvent like DMSO. the fungi i am checking is T.rubrum and M. canis.

You can use water or methanol for dissolving your plant extract, but some compounds precipitate.
In my opinion DMSO is best solvent for dissolving any plant extract and its benefits are you can store DMSO mediated stock for long time at 4 degree Celsius in refrigerator for several months.
Prepare 100 mg/ml stock of your plant extract in DMSO and further dilute it up to 10 or 20 mg/ml in water (just before analysis).

It not necessary provided your extract is soluble in the solvents. But solubility is different for different plants and different extracts. A methanol extract itself may not be completely (at least rapidly) soluble in methanol. Hence, use of DMSO is always preferred. Another factor to consider is that if you use methanol, it may have effect on its own. So the main solvent can be deionised water, or preferrably phosphate-buffered saline for cells and tissues.

actually i checked the antifungal activity of methanol itself, i didnt have much of an effect over the fungus.
