I have done asymbiotic seed germination of an orchid sp and got protocoms from them and finally seedlings. I got protocorm like bodies (PLBs) from pseudobulb segments and internodal segments taken from in vitro culture raised plants and from the fields. Can I expect somaclonal variation in Asymbiotic seed raised plants, PLBs from pseudobulb and internodal segments in in vitro culture as well as from the PLBs and plants regenerated using pseudobulb and internodal explants taken from field??
I agree with Bassam, but it only refers to seedlings. Biswajit, as I understand (according to my quite modest experience with growing orchids in vitro), you obtained PLBs from pseudobulb or internodal segments? I don't see how you can get protocorm-like bodies from segments by skipping callus phase? Please refer to:
Lee, Hsu, Yeung: Orchid protocorm-like bodies are somatic embryos. American Journal of Botany, doi: 10.3732/ajb.1300193. Therefore, somaclonal variation is expected in these cases.
