We are searching for the binding sites of all transcription factor on CMV promotor, because we are wondering if CMV promoter is activated by IFng for instance .
I do not know about your specific experiments, but I do know that in my own lab we have used the CMV promoter in many expression plasmids and YES it is regulated by several pathways. Activation of the c-Jun N-terminal Kinase (JNK) family greatly increases expression from the CMV promoter; this may also be true for other MAPKs.
We want to induce GFP under a promoter activated by NFKB, but we want to normalise by a mcherry induced by a constitutive promoteur. But, if we add TNF, or other pro-inflammatory cytokines in vitro, the CMV seems to be activated also. So it may be a problem to normalise by this mcherry if it is also increased in an inflammatory context ...
