I have a double digestion to perform and i found that the second enzyme is compatible only in cutsmart buffer. Has anyone tried double digestion with BamH1 and Xma1 and cutamart buffer?
Hi there,
According to NEBcloner, both enzymes are 100% active in CutSmart buffer.

BamH1 shows star activity according to NEB. I would like to know if anyone has performed the same. @DominiqueLiger
Hi again,
Star activity may be avoided by using HF enzymes. Also you may follow the rules from NEB if no HF enzyme is available:
https://www.neb.com/tools-and-resources/usage-guidelines/star-activity
And it works fine in my experience! Although star activity cloning could work perfectly right even before HF enzymes!!!
