We wonder what is best to use: if mimic miRNA or mimic Pre-mirna or plasmids with the miRNA to lipotranfect THP-1 PMA treated cells. We are having problems reproducing the experiments with mimics. Normally we treat the THP- cells o/n with 40nM of PMA and the following day we lipotransfect with lipofectamine 2000 with mimics at 50nM and leave them 48h followed by LPS stimulus of 8h (so cells are more than 48h without serum in OPtimem and very unhappy).

Before lipotransfection, the medium could be changed. THP-1 cells require medium change every day. After transfection, the medium can be changed again after 24-36 h without appreciably affecting the transfection outcome.
