Hi experts,

I need your advice when using Dry Transfer System for transferring proteins from gel to nitrocellulose membrane. Have you ever used home-made gels, nitrocellulose membrane, and filter papers and not the proprietary and pre-made materials from Bio-Rad?

I am used to wet transfer, which is terrific, but we want to use this Transblot Turbo Transfer System (link below) from Bio-Rad. We also want to use our own lab-made gels and lab-cut nitrocellulose membrane. The company, of course, suggest to use their own pre-made materials, however, for economical purposes, we will use our own materials.

Question: What are your protocols for using this Trans-blot Turbo using your own gels and materials (not pre-made by the company)?

a. Do you pre-wet your filter paper and nitrocellulose membrane?

b. How long is the transfer (in minutes) and what is the voltage (and other parameters) used?

c. What are the things to consider? Bad experiences?

Comparing it with wet transfer, how good is the overall transfer of proteins to the membrane using this dry transfer?
