In my deep sequencing data I have some microRNAs with zero star and loop read count but has some mature read count with significant p-value. Can I say that it's a true novel miRNA and should be reported?
you can report it as novel miRNA when you can validate its presence via wet lab techniques like for Northan blotting or RT-PCR. Only deep sequencing will not sufficient to say its presence.
you can report that you have found it but can not claim that it is amiRNA, otherwise I am sure that reviewer will ask for validation. You can say "these are are the potential miRNA and further validation is necessary to confirm it."
