If I have cultured keratinocytes expressing only K14, treated with IL-1ß, is it possible that they become activated and therefore they express K16?
That happens in basal keratinocytes, but I am not sure if "cells that express only K14 gene" means that all the other genes are not there, or they are just inhibited and eventually they could be activated).

I feel you are misunderstand my words, the genes/cells can possible to activate, it will express K16, but not effectively, due to multiple identical nature its getting hidden in analysis report. so that if its express also you unable to prove for your study, then whats the use for this?
Same way in knock out keratinocytes that not contain single system means it have lacks of same repeated, so that you can do this but you may not get useful information.,
KO keratins reactive site that means active functional analysis you look from that you can able to get point it's favor to +ve functioning, out of that if you are looking one existing active binding means you can able to understand it's functioning,

Why you have worry about this, directly you can able to culture K16 know, i feel this is best option, if you want to do this, you find the target enzyme of particular part by analyzing some proved methods, from that you get idea and do., i feel its somewhat difficult..,

Thank you for your suggestions but the thing is that I am working with K14 to study the effects of interleukin1ß on it. Therefore, I wanted to understand how it works, if genes can become activated? or if my cells only have K14 genes, that means that is impossible to have an activation and thus they cannot express K16??? I am not sure about that. And I need to understand that point. Also, in case of Knock out keratinocytes is possible that genes become activared??? or if they lack all keratins there is no possibility that genes become activated??? Thank you all. Dear Saraniya Bharathi, please could you suggest me some reference where can I read about how it works? How KO keratins or K14 can become possitve? I need to understand that point, and I have not found anywhere such explanation. Thank you very much
