Prior to selection three days after transfection  I transferred the cells from a six well to a 10 cm plate. One day later I admitted selection medium (DMEM + glutamin, 10% FCS and 10 ug/ml puromycin. Three days after selection 99.5% of the cells are dead apart from a few single cells. Has anyone also experienced this? Is the puromycin concentration too high (I had tested before on untransfected cells which concentration of puromycin would kill 100% of the cells as suggested in the protocol). Should a higher amount of plasmid increase transfection efficiency (I used 1 ug)?
